CT26.WT-luc Orthotopic Syngeneic Tumor Model of Colon Cancer: Development and Immunotherapy Response

AACR 2021 -- With clinical advancement of immunotherapy against colorectal cancer, highly translational preclinical models of the disease are required to assess the effectiveness of novel immuno-oncology agents. Orthotopic mouse tumor models grown in immunocompetent mice (i.e., syngeneic) represent a such a preclinical model with translational potential. An orthotopic model of murine colon adenocarcinoma (CT26.WT-luc) was generated using surgical methods and bioluminescence imaging (BLI) to monitor tumor progression. The sensitivity of the tumor model to immune checkpoint inhibition (targeting mouse PD-1, PD-L1 and CTLA-4) was evaluated. The tumor microenvironment of both subcutaneous and orthotopic CT26.WT-luc tumors were assessed by flow cytometry.
